The Effect of Interferon-alpha on the Foxp3^+ Regulatory T Cells in the Mice Liver Tissue of Hepatic Fibrosis

摘要 目的探讨α-干扰素(IFN-α)对四氯化碳(CCl4)诱发的肝纤维化小鼠肝组织中的Foxp3+调节性T细胞的影响及其与肝纤维化改善情况的关系。方法利用CCl4建立非病毒诱发的肝纤维化小鼠模型,造模成功后将小鼠随机分为阳性对照组、IFN-α治疗组,另设阴性对照组,每组12只。在疗程的第0、3、6、8周对小鼠肝组织做病理学检查,使用流式细胞仪分析Foxp3+调节性T细胞的百分率。结果阴性和阳性对照组的肝纤维化情况以及Foxp3+调节性T细胞百分率均无明显改变,且阴性和阳性对照组之间Foxp3+调节性T细胞百分率也无显著差异。IFN-α治疗组小鼠肝纤维化情况有明显改善(第6~8周时最为显著),且该组Foxp3+调节性T细胞百分比在治疗初期呈上升趋势,但其后出现下降,下降幅度在第6~8周最大。结论 IFN-α对Foxp3+调节性T细胞具有双向调节作用,其下调作用与小鼠肝纤维化的改善密切相关,且在第6~8周最为显著。 Objective To explore the effect of interferon-alpha on the Foxp3+ regulatory T cells in CCl4-indueed hepatic fibrosis mice dur- ing the 8-week-long IFN-α therapy,and analyze the relationship between this effect and the improvement of liver fibrosis status. Methods Tetrachloromethane (CCIa) was used to induce the liver fibrosis in mice. IFN-α therapy was administrated to the mice with liver fibrosis. At the 0,3rd, 6th, and 8th week, HE and MASSON staining were apphed to the mouse liver tissue for the histology examination, and the flow cytometry analyzer was used to obtain the percentage of the Foxp3+ regulatory T cells. Results No statistical significant difference was found on the liver fibrosis status or the Foxp3+ regulatory T cell percentages between the negative control group and the positive control group. There's no statistical significant difference of the Foxp3+ regulatory T cell percentages between the negative control group and the positive control group. The IFN-α-treated group showed an obvious improvement of the liver fibrosis status and this improvement was the most obvious during the 6th to the 8th week. Moreover, the Foxp3 + regulatory T cell percentage of the treated group went up after treatment and subse- quently declined, and the maximum rate of the decrease was observed during the 6th to the 8th week. Conclusion IFN-α has bidirectional adjusting effects on regulating the Foxp3+ regulatory T cells in CCl4-induced liver fibrosis mice. The down-regulatory effect was closely related to the improvement of the liver hbmsis status and was the most obvious during the 6th to the 8th week.
