舒必利对小鼠的吗啡所致奖赏及cAMP水平的影响

摘要 为了解多巴胺D2受体阻滞剂舒必利对吗啡奖赏特性及其与中枢cAMP水平的效应,采用条件性位置偏爱试验及放射免疫测定观察小鼠对吗啡所致奖赏的行为及中枢cAMP水平变化。结果显示:吗啡组小鼠在吗啡搭配箱体中停留时间及其中枢cAMP水平较盐水组及舒必利加吗啡组显著增加(P<0.05),而盐水组与舒必利加吗啡组之间无差异(P>0.05),说明舒必利对吗啡的奖赏特性有对抗作用,该作用是通过舒必利抑制中枢cAMP水平所致。 This study examined the effects of sulpiride, a dopamine D2 receptor antagonist, on morphine rewarding properties and its relation to cyclic AMP levels in brain. The morphine-induced reward in mice was observed in a conditione d place preference test. Cyclic AMP was estimated by radioimmunoassay. The time of staying in morphine pairing compartment and the cAMP levels in brain of mice in the morphine group increased as compared with those in other groups (P<0.05), but there was no significant difference in the time of staying in morphin e pairing compartment and the cAMP levels in brain between the saline group and the sulpiride plus morphine groups (P>0.05). These suggest that the inhibit ion of morphine-induced reward by sulpiride be due to the prevention of an incr ease in cyclic AMP levels in brain.
