摘要
作者采用戊二醛交联法将葡萄糖氧化酶固定在微孔玻璃上制成酶反 应柱,利用鲁 米诺-氯化高铁-血红素发光系统,建立了快速、简便、灵敏的固定化葡萄糖氧化酶流动注 射化 学发光法,用于血糖测定。结果显示:血清稀释1000倍后进样测定,每小时可测30个试样, 最低检出浓度达7.2×10-9 mol/L (1.3μg/L)。一个微孔玻璃柱可至少测定200次。 应用本法测定50份血样的结果与传统氧化酶比色法比较无显著性差异(两法符合程度的 相关 系数r=0.90,P>0.05)。本法取样少(10μl),操作简便、快速,酶反应柱可 用 200次以上,其准确度高(样品的加标回收率为94.0%~107.2%),因此可推广应用于大量 血样的检测。
Luminol-hemin system was used for chemiluminescent flow-injection analysis with immobilized glucoxidase(GOD) column to determine blood glucose. T he method is fast, simple and sensitive. Blood samples were diluted to 1∶1000 a n d pumped into the flow-injection analyzer for direct detection. The sampling fr e quency was 30 samples/h, with a detection limit of 7.2×10-9mol/L(1.30μg/ L). A co lumn packed with 36 milligrams of CPG could be used nearly 200 times. The determ ination results of glucose in serum by this method were in good agreement wih th ose obtained by the routine GOD spectrophotometric method (correlation coefficie nt r=0.90, P>0.05). Recoveries of this method were 94.0%~107.2%.
