摘要
目的 :探讨核心蛋白聚糖 (decorin)在肝纤维化形成机制中的作用。方法 :在正常人肝细胞株 L - 0 2及大鼠肝星形细胞株 HSC- T6体外培养体系中 ,分别加入不同质量浓度 (0 .0 1,5 ,10μg/ m l) decorin共培养不同时间后 ,进行细胞计数以及 3H- Td R和 3H- L eu掺入量测定。结果 :实验组经 0 .0 1μg/ m l decorin作用 4d或 6 d后 ,HSC- T6和 L- 0 2细胞增殖数量以及 3H- Td R和 3H- L eu掺入量均显著高于对照组 (P<0 .0 1,P<0 .0 5 )。而 5 ,10μg/ m l decorin作用 4d或 6 d后 ,HSC- T6和 L -0 2细胞增殖数量以及 3H- Td R掺入量均显著低于对照组 (P<0 .0 1,P<0 .0 5 )。 结论 :decorin对细胞体外增殖有双重调节作用 ;
Objective: To study the effects of decorin on proliferation of hepatic cells and hepatic stellate cells(HSC) in vitro . Methods: Hepatic cell(L 02) and HSC(HSC T6) were cultured with decorin at different dose. Cell number and the amount of 3H TdR and 3H Leu incorporation were determined. Results: After treatment for 4 or 6 days, decorin markedly enhanced proliferation of HSC T6 and L 02 cells at 0.01 μg/ml ( P <0.01); the amount of 3H TdR and 3H Leu incorporation were increased( P <0.05). However, decorin significantly decreased the proliferation of HSC T6 and L 02 cells ( P <0.01), the amount of 3H TdR incorporation( P <0.05) at 5 or 10 μg/ml. Conclusion: Decorin can modulate cell proliferation in vitro . It enhances cell proliferation at 0.01 μg/ml and inhibits at 5 or 10 μg/ml. Inhibition of decorin on HSC and hepatic cell proliferation can prevent liver fibrosis. [
