摘要
目的:观察隔艾炷灸对兔动脉粥样硬化(artherosclerosis,AS)中基质金属蛋白酶-2、9(matrix metallopro-teinases,MMP-2、9)表达的影响,探讨艾炷灸对兔动脉粥样硬化斑块稳定性的影响。方法:60只新西兰大耳白兔,随机分为空白组(空白对照组)、模型组(AS模型组)、艾炷灸组(AS模型加艾炷直接灸)和西药组(AS模型加阿托伐他汀),每组15只,除空白组外,其余3组通过高脂饲料喂养及免疫损伤法建立兔AS模型。运用原位杂交法检测兔动脉粥样硬化斑块中MMP-2和MMP-9mRNA的表达。结果:与模型组相比,艾炷灸组和西药组AS斑块中MMP-9mRNA的表达差异有统计学意义(P<0.05,P<0.01);西药组AS斑块中MMP-2mRNA的表达差异有统计学意义(P<0.01),而艾炷灸组AS斑块中MMP-2mRNA的表达差异无统计学意义(P>0.05)。结论:艾炷灸能改善兔AS中血管内皮MMP-9的表达。
Object: To observe the effect of direct moxibustion to mRNA expression of matrix metalloproteinases-2,9 (MMP-2, MMP-9)in artherosclerosis (AS) plaques of rabbits, to explore the effect of direct moxibustion to the stability of artherosclerosis plaques of rabbits. Method: 60 New Zealand rabbits were randomly allocated to blank, model, direct moxi- bustion, western medicine groups. 15 cases in each group. Expect blank group, the other groups were feeded by high-fat diet. The mRNA expression of MMP-2, MMP-9 in the atherosclerotic plaques of rabbits were detected by the method of in situ hy- bridization. Result:compared with model group, the expression of MMP-9 in direct moxibustion group and wesern medicine group had statistical significance( P 〈 0.05, P 〈 0.01 ) ; the expression of MMP-2 in wesern medicine group had statistical significance( P 〈 0.01 ) ; The expressions of MMP-2 in direct moxibustion group had not statistical significance( P 〉 0.05). Conclusion:direct moxibustion can reduce MMP-9 expression in the atherosclerotic rabbit.
