摘要
探讨经结核杆菌抗原特异性活化的γδT细胞的抗肿瘤活性。方法 :结核杆菌抗原 (Mtb Ag)、IL 2、PHA做为刺激原 ,活化正常人外周血单个核细胞 (PBMC) ,用流式细胞仪分析活化后淋巴细胞的表型 ,用MTT法测定活化淋巴细胞的杀伤活性。结果 :Mtb体外活化PBMC可于 18天获得大量淋巴细胞 ,且γδT细胞占有较高比例 ,对肿瘤细胞有较高杀伤活性。结论 :用Mtb为刺激原可获得大量γδT细胞 ,且在体外对肿瘤细胞有较高的杀伤活性。
Objective:To investigate the anti tumor cytotoxicity of human γδ?T cells activated by the antigen of Mycobacterium tuberculosis(Mtb). Methods:Normal human PBMC were activated by Mtb antigen and the phenotypes of proliferated lymphocytes were analyzed by flow cytometry.The in vitro cytotoxicity of activated lymphocytes against K562 were measured by the MTT assay.Results:The proliferation rate of lymphocytes activated by Mtb antigen and cultured in IL 2 for 18 days were about 16 fold,in which the proportion of γδ?T cells were about 60%.The activated lymphocytes stimulated by Mtb antigen were detected to have cytotoxicity in vitro against tumor cell K562.Conclusions:The high propotion of γδ?T cells can be gained in human PBMC activated by Mtb antigen,and the activated lymphocytes have high cytotoxicity in vitro against tumor cells.
