糖尿病大鼠红细胞山梨醇浓度与肾脏损害的相关性

摘要 目的研究糖尿病大鼠红细胞山梨醇浓度与肾脏损害的相关性。方法链脲佐菌素52 mg/kg一次性腹腔注射大鼠,造模,于不同的时间点测定糖尿病大鼠红细胞内山梨醇、血肌酐、24 h尿量、尿素氮、及尿肌酐,并计算肌酐清除率,观察肾脏的病理改变,研究红细胞山梨醇浓度与肾脏损害的相关性。结果糖尿病大鼠红细胞内山梨醇由第3周开始浓度显著升高,第6周后趋于稳定;肾功能和肾脏的病理改变则在第6周后变化明显。结论糖尿病大鼠红细胞内山梨醇聚集达一定浓度,并持续一段时间后糖尿病大鼠出现肾脏损害,红细胞内山梨醇浓度可作为肾脏损害前的预警指标。 Objective To study the correlation of erythrocyte sorbitol concentration and kidney injury in diabetic rats. Methods Diabetic rat model was created by intraperitoneal injection of streptozocin(52 mg/kg). The kidney pathological changes were observed through the measurement of erythrocyte sorbitol concentration, the plasma concentration of creatinine, 24 h urine volume and the concentrations of urea nitrogen and creatinine in urine at different time, and the calculation of creatinine clearance rate. The correlation between erythrocyte sorbitol concentration and kidney injury was investigated. Results The erythrocyte sorbitol concentration was gradually increased from the third week, and significantly increased at the sixth week, then tended to be stable in diabetic rats. Renal function and kidney pathological changes were obviously altered after 6 weeks. Conclusion The erythrocyte sorbitol concentration in diabetic rats accumulates to a certain degree and lasts for a period of time, then the kidney injury appears. Hence, the erythrocyte sorbitol concentration can be served as the early-warming index of kidney injury.
