Improvement on the Key Synthesis Process of Progesterone

摘要 研究了以TEMPO为催化剂、妊娠烯醇酮为原料,经次氯酸钠氧化得到目标产物黄体酮的新工艺,考察了该工艺中氧化剂、投料比、反应体系pH、反应温度等对反应收率和产品质量的影响。结果表明,以次氯酸钠为氧化剂,妊娠烯醇酮、次氯酸钠、TEMPO、溴化钠的投料摩尔比为10:15:0.8:0.5,pH为9.3,反应温度为0~5℃时,总收率达到63%,产品纯度较高,具有工业化前景。 A new technology process of target product progesterone oxidized by sodium hypochlorite with TEMPO as catalyst and pregnenolone as raw material was researched.The effects of oxidant,raw materials molar ratio,pH of reaction system and reaction temperature on the yields and product quality were discussed.The results showed that the better oxidant was sodium hypochlorite,the optimal molar ratio of pregnenolone,sodium hypochlorite,TEMPO,sodium bromide was 10:15:0.8:0.5,the better pH of reaction system was about 9.3,the reaction temperature should be controlled at 0~5 ℃.The overall yield of this process was up to 63% and the products had high purity which has prospects for large production.
