摘要
目的探讨青蒿素(artemisinin)对人乳腺癌细胞血管内皮生长因子(VEGF)表达的影响及作用机制。方法应用四甲基偶氮唑盐(MTT)法检测青蒿素对人乳腺癌细胞系MCF-7和MDA-MB-231细胞增殖的影响,应用酶联免疫吸附试验(ELISA)检测VGEF蛋白的分泌水平,采用半定量逆转录-聚合酶链反应(RT-PCR)技术检测VEGF mRNA表达水平。结果青蒿素对MCF-7和MDA-MB-231细胞均有明显增殖抑制效应。青蒿素降低了VEGF分泌水平和VEGFmRNA的表达水平,并呈剂量-时间效应关系。结论青蒿素能抑制人类乳腺癌细胞增殖和细胞VEGF mRNA的表达。
Objective To assess the effects of artemisinin on the VEGF mRNA level in human breast cancer cell lines. Methods MDA-MI3-231 cells and MCF-7 cells were used as the study objects. The MTT assay was used to detect the effect of artemisinin on the cells viability. ELISA was used to determine the protein level of VEGF. Breast cancer cells were treated with art^nisinin for 24-72 h, and RT-PCR was used to detect the level of VEGF mRNA expression. Results Artemisinin had visible inhibitory effect on cell viability. Artemisinin reduced the secretion of VEGF and inhibited the level of VEGF mRNA expression. Conclusions Artemisinin can inhibit cell viability and the level of VEGF mRNA expression in breast cancer cells.
