摘要
目的探讨FAT10过表达对肝癌细胞HepG2侵袭及迁移的影响。方法采用脂质体转染法建立FAT10基因过表达的HepG2细胞株;采用蛋白质印迹法检测转染重组表达载体pcDNA3-FAT10后,HepG2细胞中FAT10蛋白的表达水平。体外侵袭实验及划痕实验检测FAT10基因过表达对HepG2细胞迁移及侵袭能力的改变,并采用芯片检测FAT10下游与细胞侵袭转移相关基因的差异表达。结果蛋白质印迹法检测结果显示,转染重组表达载体pcDNA3.1-FAT10的HepG2细胞中FAT10蛋白均过表达;FAT10过表达能明显提高HepG2细胞的侵袭及迁移能力。结论 FAT10基因在HepG2细胞中过表达能提高肝癌的侵袭及迁移能力,并促进肝癌的发生及发展。
Objective To investigate the effects of FAT10 over-expression on cell invasion and migration of human liver carcinoma hepG2 cells in vitro.Methods The FAT10-over-expressing HepG2 cells were constructed using Lipofectamine 2000.The expression levels of FAT10 protein in HepG2 cells transfected with pcDNA3-FAT10 were detected by Western blotting.The migration and invasive capabilities of HepG2 cells were detected by scratch-wound migration assay and transwell invasion assay,respectively.The differential expression of genes related to cell migration and invasion in the downstream of FAT10 was determined by gene chip.Results Western blotting revealed that FAT10 protein in the HepG2 cells transfected with pcDNA3-FAT10 was over-expressed.The invasive and migration capabilities of HepG2 cells transfected with pcDNA3-FAT10 were significantly increased.Conclusions FAT10 over-expression plays an important role in the invasion and migration of human hepatocellular carcinoma HepG2 cells.It can promote the progression of human hepatocellular carcinoma.
