高效液相色谱法测定益肾颗粒中淫羊藿苷的含量

Determination of icariin content in Yishen granules by HPLC

[目的]建立益肾颗粒中淫羊藿苷含量的测定方法。[方法]采用高效液相色谱法(HPLC),色谱柱为美国Waters公司C18柱(250 mm×4.6 mm,10μm),流动相为乙腈-0.1%磷酸溶液(28∶72),流速为1 mL/min,检测波长为270 nm,柱温为室温。[结果]淫羊藿苷在0.048 5-0.970 0μg内与色谱峰面积线性关系良好(r=0.999 9),平均回收率97.0%,RSD=0.7%(n=6)。[结论]该法简便、专属性、重复性好,可用于益肾颗粒中淫羊藿苷的含量测定。

[Objective] To establish a method to determine the content of icariin in Yishen granules. [Methods] Using the method of HPLC, the chromatographic column was the United States Waters C18 column （250 mm x 4.6 mm, 10 p^m）. The mobile phase was methyl cyanides-0.1% phosphoric acid solution （28：72）. Velocity of flow rate was 1 mL/min. The detection wavelength was 270 nm, and column temperature was at room temperature. [Results] Icariin in 0.048 5 ~0.970 0 I^g and chromatographic peak area had good linear relationship （r=0.999 9）. The average recovery was 97%, RSD=0.7% （n=6）. [Conclusion] The method is simple, specific, reproducible, and can be used for determination of ieariin in Yishen granules.
