摘要
研究新城疫病毒 (NewscastlediseaseVirus,NDV)强、弱毒株在不同的传代细胞的生物学特征。F4 8E8强毒株在BHK、Vero和CEF中都能增殖并产生细胞病变 ,而V4生弱毒株不能在BHK和Vero中增殖。当MEM含 1 0ug/mL胰蛋白酶时 ,NDV弱毒株在BHK和Vero中能较好地复制 ,将V4株在Vero中传代培养后 ,Vero中传代的V4毒株的HA较低 ,但毒力增强 ,BHK中传代的V4毒株的毒力和HA都提高 ,能达到其鸡胚毒上的毒力和HA效价。如果进一步研究能够证实在BHK上适应增殖的V4毒株的免疫原性比较好 ,那么就可能用BHK细胞系替代鸡胚增殖V4株作为疫苗的病毒抗原。使用BHK等传代细胞制备的NDV疫苗具有避免传统疫苗散播其他疾病的危险、降低成本和大规模生产等优点。使用传代细胞代替鸡胚来生产NDV疫苗 。
The experiment was taken in order to study the biological characteristics of Newscastle Disease Virus(NDV) strong and weak strain on BHK cells. The results showed that F 48 E 8 could proliferate in BHK cell,Vero cell and CEF cell, and make the cells pathological changes, but the V4 couldn't proliferate in BHK cell and Vero cell. When the concentration of trypsinogen is 10ug/ml in MEM, weak NDV strain could copy in BHK cell and Vero cell. Cultivated V4 strain in Vero cell, the HA of V4 in Vero cell was lower, virulence became strong, the HA and virulence of V4 in BHK cell were both increase , equaled to the virulence and HA on chicken embryo. It would be approved that immunogen of V4 proliferated in BHK cell was better after further study, it is possible use BHK cell series to replace chicken embryo to proliferate V4 as virus antigen of vaccine.
