PAPS免疫微球快速诊断猪旋毛虫病的研究 被引量:1

THE APPLICATION OF PAPS IMMUNOSPHERE FOR THE DETECTION OF SWINE TRICHINELLOSIS

摘要 应用新型的聚醛化聚苯乙烯 ( PAPS)载体微球与最佳的旋毛虫抗原共价交联制备成特异性强、敏感性高、重复性好的快速诊断试剂 ,应用于猪旋毛虫病的生前诊断。我们对旋毛虫各个发育期的虫体和分泌排泄抗原进行了分析研究 ,并以同一蛋白质浓度 ( 0 .6mg/m L)的成虫、新生幼虫、肌幼虫、成虫 ES、肌幼虫 ES、肌幼虫 A峰、肌幼虫 B峰等七种抗原分别与 PAPS载体微球共价交联制成各种快诊试剂 ,然后进行效果比较试验 ,其结果是肌幼虫 ES抗原最佳 ,其次是干燥冷浸肌幼虫抗原和肌幼虫 A峰抗原。其它抗原都呈现不同程度的非特异性反应。应用干燥冷浸肌幼虫抗原制成的 PAPS快诊试剂对 30头人工感染猪的阳性检出率为 1 0 0 %,对 1 0 0头上海非流行区猪试验结果是阴性符合率为 98%,对流行区 (河南南阳、湖北襄樊、云南大理 ) 2 97头猪检测结果是阳性反应 30头 ,而 ELISA法阳性反应 2 9头 ,结果相近。本法敏感、特异、快速、简易 。 The suitable Trichinella spiralis antigens are covalently linked with Polyaldehyde Polystyrene Immunosphere (PAPS).The specific,sensitive,reproducible quick diagnostic reagents are used for the detection of swine Trichinellosis.PAPS is linked at the same concentration (0.6mg/mL) with adult,new born larvae,muscle larvae,adult ES,muscle larvae ES,peak A and peak B of muscle larvae antigens,Comparison show muscle larvae ES is best, and dried soaked muscle larvae antigen and peak A antigens are also good,though others appear non specific reaction,The test using PAPS quick diagnostic reagents linked with dried soaked muscle larvae antigen show that when detecting swine infected by Trichinella spiralis, positive rate is 100%,when detecting swine from non epidemic zone,negative coincidence rate is 98%,and when detecting 297 swine from epidemic zone,30 swine have positive reaction,similar to the result of ELISA,by which 29 swine do.The method is sensitive,specific,quick,simple and suitable for food health quarantine and anti epidmic quarantine.
