摘要
应用旋毛虫肌幼虫干燥冷浸可溶性抗原与聚醛化聚苯乙烯 ( PAPS)载体微球共价交联制成快诊试剂 ,对旋毛虫感染兔血清进行检测 ,初步探明其血清抗体的消长规律。实验显示 ,以1 60 0 0条肌幼虫 /只经口饲喂感染 1 0只兔 ,于第 7天可检出抗体 ,并分别于感染后第 45~ 49天先后达到抗体最高水平 ,然后维持一段时间 ( 2 0~ 30天左右 ) ,以后逐渐缓缓下降。此结果与旋毛虫肌幼虫感染宿主后的生活周期相吻合。肌幼虫在肠道内发育为成虫 ,雌虫于感染后 7~ 1 0天产幼虫 ,幼虫转入体循环 ,并进入横纹肌纤维 ;从第 3周开始形成包囊 ,到第 7~ 8周完成。幼虫进入肌纤维及开始成囊是机体产生免疫应答最强的时期 ,可产生高水平抗体并维持一段时间 ;随着成囊完成、肌幼虫钙化 ,刺激兔体产生相应抗体的能力越来越弱 ,血清抗体滴度缓缓降低。由此可见 ,制备阳性血清 ,应以感染后第 49天至
Trichinella spiralis dried and soaked muscle larvae antigen is covalently linked with polyadehyde polystyrene immunosphere (PAPS),and this quick diagnostic reagent is used to detect the sera antibody titer of rabbits fed with 16 000 T.Spiralis muscle larvae.The antibody could be detected by day 7 of infection, and the antibody titers reach highest point by day 45 to 49,and keep up for about 20 to 30 days,then decline gradually and slowly.The result accords with the life cycle of T.spiralis in host.Larvae are born on 7~10th day then get into blood circulation and enter striated muscle.By week 3 of infection encystment begins and finishes by week 7~8. When larvae enter striated muscle and encystment begins,hosts generate strongest immune response,and produce high titer antibody and keep it up. While encystment finished and muscle larvae calcified,rabbits immune response become weaker and weeker,and antibody titers decline gradually.Hence proper time for producing positive sera is 49~60th day after infection.
