HPV-E7联合淋巴毒素免疫诱导的生殖道特异性免疫应答

Specific immune responses of genital tract induced by intranasal immunization with HPV-E7 and lymphotoxin

摘要 目的鼻腔免疫小鼠,诱导生殖道局部黏膜的特异性免疫应答。方法将18只小鼠随机均分为三组:A组、B组小鼠鼻腔滴注人乳头瘤病毒(HPV)E7 10μg,B组静脉注射淋巴毒素(LT)0.05μg;C组用PBS作为对照。收集小鼠阴道冲洗液,ELISA测定HPV-E7特异性阴道IgA抗体滴度。在体内细胞毒性T淋巴细胞(CTL)活性分析实验中,用流式细胞仪检测靶细胞清除率。结果 A组阴道冲洗液中检测到HPV-E7特异性抗体滴度约为1∶20,高于B组的1∶40(P<0.05)。A组细胞杀伤率为(48.64±3.07)%,低于B组的(69.12±9.01)%(P<0.05)。结论HPV-E7经鼻腔免疫小鼠后,可以使生殖道局部黏膜产生特异性体液和细胞免疫应答;LT则有效地提高了生殖道局部黏膜免疫应答效应。 Objective To explore the specific immune responses of genital tract induced by intranasal immunization with human papillomavirus-E7 ( HPV-E7 ) and lymphotoxin (LT). Methods Eightee mice were equally randomized into three groups of A (immunized with HPV-E7 ), B (immunized with HPV-E7 plus LT) and C(PBS control). The HPV-E7-specific vaginal IgA in the vaginal washing fluid was detected by ELISA. The response of cellular immunity was evaluated by measuring the clearance of target cells with flow cytometry in vivo cytotoxicity assay. Results The titer of HPV-E7-specific antibody was higher in group A than that in group B(1: 20 vs. 1: 40) (P〈0. 05). The cell-killing rate was lower in group A than that in group BE (48. 64 ± 3.07)% vs. (69. 12±9.01)%](P〈0. 05). Conclusion Specific humoral and cellular immune responses of genital tract are induced by intranasal immunization with HPV-E7, which can be effectively promoted by LT.
