类风湿性关节炎患者滑膜组织中β1,4-半乳糖基转移酶-Ⅰ的表达 被引量:3

β1,4-Galactosyltransferase-Ⅰ expression in synovial tissues of patients with rheumatoid arthritis

摘要 目的研究β1,4-半乳糖基转移酶-Ⅰ(β1,4-GalT-Ⅰ)在类风湿性关节炎(RA)患者膝关节滑膜组织中表达水平及细胞定位。方法选择12例RA患者(A组)和8例健康人(B组)滑膜组织,采用Western blot检测β1,4-GalT-Ⅰ、E-选择素的蛋白表达水平,免疫组化检测β1,4-GalT-Ⅰ的表达和分布情况,免疫荧光检测β1,4-GalT-Ⅰ与滑膜组织中炎症细胞以及E-选择素的共定位情况。结果与B组相比,A组滑膜组织中β1,4-GalT-Ⅰ和E-选择素蛋白表达明显增加(P<0.01或P<0.05)。在RA滑膜炎症过程中,β1,4-GalT-Ⅰ主要表达于巨噬样滑膜细胞、成纤维样滑膜细胞、活性T细胞和中性粒细胞。E-选择素与β1,4-GalT-Ⅰ存在共定位。结论β1,4-GalT-Ⅰ可能参与调节RA滑膜组织的炎症反应过程。 Objective To investigate the expression and location of β1,4-galactosyltransferase- Ⅰ (β1,4-GalT-Ⅰ ) in synovial tissues of the knee joint from patients with rheumatoid arthritis (RA). Methods The synovial tissue samples were taken from 12 RA patients(group A) and 8 healthy people(group B). The protein expressions of β1,4-GalT-Ⅰ and E-selectin were detected by Western blot. The expression and distribution of β1,4-GalT-Ⅰ were determined by immunohistochemistry. The cellular colocalization of β1,4-GalT-Ⅰ with inflammatory cells and E-selectin was observed by double immunofluorescence staining. Results Compared with group B, the protein expressions of β1,4-GalT-Ⅰ and E-selectin in the synovial tissues were significantly increased in group A(P〈0. 01 or P〈0.05). β1,4-GalT-Ⅰ was mainly expressed in macrophageqike synoviocytes, fibroblast-like synoviocytes, active T cells and neutrophils. Moreover, E-selectin was colocalized with β1,4-GalT-Ⅰ in the synovial tissues. Conclusion β1,4-GalT-Ⅰ may play an important role in the inflammatory reaction in the synovial tissues of RA oatients.
