摘要
目的:制备并鉴定苏丹红Ⅳ和柠檬黄人工抗原。方法:琥珀酸酐法构建苏丹红Ⅳ半抗原;以牛血清蛋白(BSA)为载体,用活性脂法分别与苏丹红Ⅳ半抗原和柠檬黄进行偶联。并用紫外吸收色谱法和电泳凝胶色谱鉴定制备是否成功。结果:经紫外吸收和摩尔吸收系数法估算得到苏丹红Ⅳ与BSA的偶联比31∶1,柠檬黄与BSA的偶联比17∶1。结论:成功制备出苏丹红Ⅳ和柠檬黄的人工抗原。
Objective:To prepare and identify the artificial antigen for Sudan Ⅳ and tartrazine.Methods: The Sudan Ⅳ hapten was prepared by succinic anhydride with Sudan Ⅳ.Bovine serum albumin(BSA) was served as the carrier protein to couple with the Sudan Ⅳand tartrazine hapten,respectively.The complete antigens were identificated by ultra-violet spectrophotometry and sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE).Results: The results from UV spectral and molar absorbance coefficient showed the coupling ratio of Sudan Ⅳ hapten to BSA was 31,while that was only 17 for tartarzine-BSA conjugate.Conclusion: The immunogen based on the Sudan Ⅳ hapten and tartrazine was synthesized successfully.
