摘要
目的研究活血化瘀通络中药对糖尿病胃轻瘫大鼠的作用及可能机制。方法雄性SD大鼠随机分为对照组、模型组、中药1组(丹参、川芎组)、中药2组(水蛭、地龙组)、中药3组(丹参、川芎与水蛭、地龙合方组)。除对照组外,其余各组给予链脲佐菌素(STZ)(60 mg/kg)腹腔注射制备糖尿病大鼠模型。中药1组、中药2组、中药3组大鼠分别给予中药1.2 g/(kg.d)灌胃,对照组、模型组给予等容积0.9%氯化钠注射液灌胃。于用药后第6周末杀检动物,免疫组织化学法测定胃肌层组织c-Kit和间隙连接蛋白43(Cx43)的灰度值。结果模型组、中药1组、中药2组、中药3组与对照组比较c-Kit阳性细胞分布密度明显减弱(P<0.05);中药1组、中药2组、中药3组与模型组比较c-Kit阳性细胞分布密度增强(P<0.05);中药3组与中药1组、中药2组比较c-Kit阳性细胞分布密度增强(P<0.05)。模型组、中药1组、中药2组、中药3组与对照组比较Cx43的表达明显减低(P<0.05);中药1组、中药2组、中药3组与模型组比较Cx43的表达升高(P<0.05);中药3组与中药1组、中药2组比较Cx43的表达升高(P<0.05)。结论活血化瘀通络中药可能通过调控Cajal间质细胞(ICC)和Cx43的阳性表达促进糖尿病胃轻瘫大鼠的胃排空。
Objective To study the effect and mechanism of herbs for activating blood,dissolving stasis and dredging collaterals on interstitial cells of Cajal and connexin 43 in rats with diabetic gastroparesis.Methods SD male rats were randomly divided into 5 groups;control group,model group,Chinese medicine group one(Salvia miltiorrhiza and Rhizoma Chuanxiong),Chinese medicine group two(Leeches and Earthworm) and Chinese medicine group three(Salvia miltiorrhiza,Rhizoma Chuanxiong,Leeches and Earthworm).SD male rats were injected streptozotocin(STZ) solution(60mg/kg) into abdominal cavities for diabetic melliutus rat model except for control group.The rats in Chinese medicine group one,Chinese medicine group two and Chinese medicine group three were given by intragastric administration with the Herbs respectively 1.2 g/(Kg·d),control group,model group were given equal volume of normal saline.After 6 weeks,interstitial cells of Cajal and connexin 43 gray value was detected by immunohistochemical method(s-p).Results The c-Kit positive cells distribution density in model group,Chinese medicine group one,Chinese medicine group two and Chinese medicine group three were decreased compared with that in control group(P〈0.05).The c-Kit positive cells distribution density in Chinese medicine group one,Chinese medicine group two and Chinese medicine group three were increased compared with that in model group(P〈0.05).The c-Kit positive cells distribution density in Chinese medicine group three were increased compared with that in Chinese medicine group one and Chinese medicine group two(P〈0.05).The Cx43 gray value in model group,Chinese medicine group one,Chinese medicine group two and Chinese medicine group three were lower than that in model group(P〈0.05).The Cx43 gray value in Chinese medicine group one,Chinese medicine group two and Chinese medicine group three were increased compared with that in model group(P〈0.05).The Cx43 gray value in Chinese medicine group three were increased compared with that in Chinese medicine group one and Chinese medicine group two(P〈0.05).Conclusion Herbs for activating blood,dissolving stasis and dredging collaterals can improve the gastric emptying of rats with diabetic gastroparesis by regulating interstitial cells of Cajal and connexin 43 expression.
