UV法和HPLC法测定藏药多腺悬钩子中总黄酮及芦丁的含量

Determination of Total Flavonoids and Rutin Contents of Rubus phoenicolasius by UV and HPLC

对藏药多腺悬钩子(Rubus phoenicolasius)中总黄酮和芦丁含量分别进行了测定。采用UV法测定总黄酮的含量,总黄酮在0.20～1.00 mg(R2=0.999 6)时线性关系良好,平均回收率为99.2%。采用HPLC法测定芦丁的含量,使用的色谱柱为Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm),柱温30℃,流动相为甲醇∶0.4%磷酸溶液(45∶55,V/V),结果表明,芦丁在0.02～0.10μg(R2=0.999 8)时线性关系良好,平均回收率为102.6%。该方法简单、可靠、快速。

The contents of total flavonoids and rutin of Rubus phoenicolasius were determined. Total flavonoids were deter- mined by ultraviolet spectrophotometry （UV）. Results showed the standard curve of total flavonoids was linear in the range of 0.20-1.00 mg （R^2=0.999 6） and its average recovery ratio was 99.2%. Rutin was determined by high performance liquid chromatography （HPLC）, the column was Agilent Eclipse XDB-C18 （4.6 min×250 mm,5 μm）, the column temperature was 30 ℃, and the sample was separated with methanol and water containing 0.4% phosphpric acid solution （45：55,V/V）. The stan- dard curve of the rutin was linear in the range of 0.02-0.10μg （R^2-0.999 8） and the average recovery was 102.6%. The method is easy,rapid and reliable.
