Optimum of in vitro Regeneration System of Chinese Kale

摘要 以芥蓝(Brassica oleracea)真叶和子叶段为外植体,研究了TDZ浓度、基因型及苗龄等因素对芥蓝不定芽再生的影响。结果表明,真叶和子叶段的最佳不定芽诱导培养基分别为MS+0.50~1.00 mg/L TDZ和MS+0.50 mg/L TDZ,真叶和子叶段再生频率分别达到82.56%和89.06%。最佳生根培养基为1/2 MS+0.10 mg/L NAA,其生根率达到100.00%,再生植株移栽成活率为96.00%。 Leaves and cotyledonary segments were used as explants of Chinese kale (Brassica oleracea), the effects of TDZ concentration, genotypes and seedling age on regeneration shoots of Chinese kale were studied. The results showed that the optimum culture medium of leaves and cotyledonary segments to induce buds were MS+0.50-1.00 mg/L TDZ arid MS+0.50 mg/L TDZ respectively. The maximum frequency of leaves and cotyledonary segments shoot regeneration was 82.56% and 89.06%. The best medium for roots differentiation was 1/2 MS+0.10 mg/L NAA, and the rooting rate was up to 100.00%. After acclimatization, the plantlets were transplanted to pots with nutritious soil, the survival rate was up to 96.00%.
