摘要
为了给西南桦种苗繁育与倍性育种体系的建立提供参考,以西南桦种子无菌萌发苗的顶芽和带叶茎段外植体为材料,采用组织培养方法建立芽增殖快繁体系。结果表明:在3/4MS+2.5%蔗糖+0.65%琼脂+肌醇0.1mg/L的芽诱导基本培养基中,添加浓度为0.5mg/L的BA对顶芽的增殖效果最好,芽增殖系数为2.57倍,添加BA 0.5mg/L+NAA0.01mg/L进行西南桦顶芽和带叶茎段芽增殖培养的效果最佳,芽增殖系数为3.32倍和1.62倍;西南桦无菌培养苗以90d苗龄最适合作为顶芽和带叶茎段外植体进行增殖培养。
The apical buds and stem segments from B. alnoides sterile seedlings were cultured to establish a rapid propagation system of bud proliferation for its seedling propagation and ploidy breeding. The results showed that the bud proliferation coefficient of the apical buds cultured on 3/4 MS +2.5 sucrose + 0. 650//oo agar 4-0.1 mg/L inositol 4- 0.5 mg/L BA medium was 2.57 and the bud proliferation coefficient of the apical buds and stem segments with two leaves cultured on 3/4 MS 4- 2.5 % sucrose 4- 0.65 Gagar 4-0. 1 mg/L inositol 4- 0. 5 mg/L BA 4- 0. 01 mg/L NAA medium was 3. 32 and 1. 62 respectively. The optimum seedling age for proliferation culture of the apical bud and stem segment with two leaves was 90 d.
