苦参碱诱导K562白血病细胞分化和凋亡的实验研究 被引量:118

摘要 研究苦参碱的抗肿瘤作用。方法 :以人红白血病细胞株K562为研究对象,通过形态学观察和细胞化学染色了解不同浓度的苦参碱对肿瘤细胞的诱导分化作用,并利用程序性细胞死亡检测试剂盒检测肿瘤细胞凋亡的情况。结果 :0 1g/L苦参碱能够诱导K562白血病细胞分化,形态学上类似中幼红和晚幼红细胞,联苯胺染色阳性率由1 %~2 %上升至7 %,细胞化学染色显示糖原由阴性转为强阳性且阳性率为100 %;1 0g/L苦参碱作用24h,30 %的K562细胞可出现明显的空泡,并有凋亡小体出现,末端标记法也检测到凋亡的出现。结论 :0 1g/L苦参碱能够诱导肿瘤细胞的分化,1 Objective: To investigate the anti tumor effect of matrine, a main composition of Sophora flavescens Ait Methods:Differentiation ofhuman erythroleukemia K562 cells was assessed by morphological changes and cytochemical stain, while the apoptosis was demonstrated by programmed cell death (PCD) assay Results: K562 cells treated with 0 1 g/L matrine were found differentiated and morphologically changed to polychromatic normoblasts and orthrochromatic normoblasts Treatment of the cells with matrine increased the percentage of benizidine positive cells from 1%~2%to 7% Cytochemical stain demonstrated that the periodict acid schiff (PAS) changed from negative to strong positive to give the positive rate of 100% Treatment of the cells with 1 0 g/L matrine for 24 hours induced distinct vacuoles and some apoptotic bodies in 30%of the cells, and apoptosis were also demonstrated by PCD Conclusion: Matrine in concentration of 0 1g/L can induce K562 cells differentiation while in concentration of 1 0g/L will induce apoptosis
