摘要
根据已知的抗菌肽moricin核苷酸和氨基酸序列设计兼并引物,利用PCR方法克隆得到柞蚕新型抗菌肽moricin.该序列长度为126bp,将该序列克隆到pUCm-T载体.通过测序鉴定了柞蚕moricin的核苷酸序列,其核苷酸序列与家蚕moricin II的相似性为99.21%.根据核苷酸序列推算的柞蚕氨基酸序列与家蚕moricin II和mori-cin I的相似性分别为97.62%和95.24%.无论柞蚕moricin的核苷酸还是氨基酸序列都与家蚕moricin相似,说明它们属于同一抗菌肽家族,柞蚕核苷酸序列已在美国国立生物技术信息中心登录,登录号为DQ519400.
To obtain moricin from Antherea pernyi,PCR was performed.Degenerate primers were designed based on the conserved amino acid sequences of moricin.A 126 bp cDNA fragment was obtained from Antherea pernyi,which encodes a peptide of 43 amino acid residues,and was cloned into pUCm-T vector.The nucleotide sequence of the moricin was confirmed by sequencing.Nucleotide sequences of Antherea pernyi moricin showed 99.21% homology with Bombyx mori moricin II,the deduced amino acid sequence showed 97.62% and 95.24% homology with Bombyx mori moricin II and moricin I.Not only nucleotide sequences but also amino acid sequence of moricin from Antherea pernyi like that of moriciin from Bombyx mori,confirming that moricin from Antherea pernyi is a new moricinlike antibacterial peptide.This sequence was deposited in national center for biotechnology information GenBank with the Accession No.DQ519400.
