摘要 采用2.5%右旋葡聚糖硫酸钠(DSS)诱导小鼠溃疡性结肠炎模型,研究Lactobacillus casei-14(L.casei-14)对小鼠溃疡性结肠炎(UC)的预防作用,并探讨其可能的机制.结果表明:L.casei-14高剂量组小鼠结肠长度为(7.66±0.58)cm,与模型组((6.73±0.42)cm)相比差异显著(P〈0.05);中、高剂量组小鼠结肠炎疾病活动指数(DAI)分别为(4.22±0.97)和(4.17±1.33),与模型组(8.00±2.12)相比差异极显著(P〈0.01);组织病理学炎症评分结果显示,中、高剂量组小鼠炎症评分分别为(4.88±0.83)和(4.13±0.64),极显著低于模型组(10.50±1.85)(P〈0.01);中、高剂量组小鼠IL-1β、TNF-αmRNA表达水平均显著低于模型组,与正常对照组没有极显著性差异(P〈0.01).说明L.casei-14对小鼠溃疡性结肠炎具有预防作用,其机制可能是通过抑制IL-1β和TNF-α炎症细胞因子的表达而减轻小鼠结肠黏膜的损伤,以达到预防UC的目的. The model of UC was induced with 2.5% of dextran sulfate sodium(DSS) in mice,the preventive effects of Lactobacillus casei-14(L.casei-14) on ulcerative colitis(UC) were studied,and the possible mechanism was discussed.The results showed that compared with the model group((6.73 ± 0.42)cm),the colon length was significantly increased(P0.05) in L.casei-14 high-dose group((7.66±0.58)cm).DAI of medium-and high-dose groups were(4.22±0.97) and(4.17±1.33) respectively,and showed a contrasting significant reduction compared with the model group(8.00±2.12)(P0.01);Histopathological score of medium-and high-dose groups were(4.88±0.83) and(4.13±0.64) respectively,and significantly reduced compared with the model group(10.5±1.85)(P0.01).The expression levels of IL-1β,TNF-α mRNA in medium-and high-dose groups were significantly lower than that in the model control group.There was no remarkable difference compared with the normal group(P0.01).L.casei-14 showed preventive effects on ulcerative colitis,and reduced the damage of colonic mucosa in mice.The mechanism was probably through inhibiting the expression of inflammation cytokines of IL-1β and TNF-α and to prevent or control UC.
