油松苯丙氨酸解氨酶基因的克隆与序列分析 被引量:3

摘要 利用同源克隆的方法,从油松(Pinus tabulaeformis)中克隆得到苯丙氨酸解氨酶(PAL)基因的cDNA和DNA序列,cDNA序列全长2 157bp,编码718个氨基酸,包含有完整的ORF,命名为TaPAL(GenBank登录号:JX280460)。通过核苷酸序列多重比较表明,TaPAL基因与其他植物PAL基因高度同源,预测的蛋白序列包含与水稻、玉米PAL相同的脱氨基位点和催化活性位点。PAL系统进化树显示,TaPAL与松科类植物PAL聚类关系最近。另外,对比发现TaPAL基因组DNA序列与cDNA序列完全一样,表明该基因无内含子。 The cDNA and DNA sequence of phenylalanine ammonia-lyase gene was cloned from Pinus tabulaeformis for the first time by the method of homology cloning.The full length of cDNA was 2 157 bp,containing a complete open reading frame and 718 amino acids were encoded,designated as TaPAL(Accession number:JX280460).It was indicated that the nucleic acid sequence of TaPAL shared a high identity with other plants' via multiple alignment.The domination sites and catalytic active sites appeared in PAL protein of Oryza and Zea mays were also found in that of inferred protein sequence.Phylogenetic tree analysis suggested that TaPAL was much closer to PALs from Pinacease than that from other plants.Moreover,this gene was found to be intronless because the cDNA and genomic DNA sequence of TaPAL were completely the same.
