RP215单抗的^(131)I标记及其对裸鼠肿瘤的作用

摘要 采用Iodogen固相法进行131I标记单克隆抗体RP215。结果显示,131I-RP215标记率为68.3%,放射化学纯度为98.5%,比活度为30.3 GBq/g,标记物在-20℃存放5 d后放化纯度降至85.6%。进行了131I标记的RP215单克隆抗体对肿瘤抑制作用研究。结果表明131I-RP215能较强地抑制小鼠体内移植肿瘤的生长,131I-RP215在治疗实体瘤中有潜在的应用前景。 Monoelonal antibody RP215 was labeled with 131I by Iodogen method. The labeling efficiency was 68.3% , the radioehemieal purity was 98.5% , while the specific activity was 30. 3 GBq/g. The radioehemieal purity of 131I - RP215 reduced to 85.6% after 5 clays in vitro storage( -20 ℃ ). Antitumor effect of 131I - RP215 was studied. The results suggested that 131I -RP215 had potential prospect of clinical application in the treatment of solid tumor such as ovarian carcinoma.
