响应面法优化微胶囊固定化粘红酵母生产L-苯丙氨酸的工艺条件

Optimization of Conditions for Producing L-Phenylalanine by Rhodotorula Glutinis Immobilized in Microcapsule Using Response Surface Methodology

摘要 以微胶囊固定化粘红酵母为研究对象,考察了粘红酵母转化反式肉桂酸(t-CA)生成L-苯丙氨酸过程中转化体系温度、pH值、cNH+4/ct-CA对L-苯丙氨酸积累量的影响,并通过响应面法对转化条件进行了优化,得到最佳转化条件为:温度30.75℃、pH值11.58、cNH+4=4.77mol.L-1、ct-CA=10g·L-1,在该条件下,L-苯丙氨酸积累量为27.5g·L-1。 Taking Rhodotorula glutinis immobilized in mierocapsule as the research object, the effects of temperature,pH value,NH+ 4concentration and t-CA concentration on L-Phe yield using t-CA as substrate by Rhodotorula glutinis were investigated. The parameters of reaction were optimized by response surface method- ology. The optimum conditions were obtained as follows:temperature was 30.75 ℃ ,pH value was 11.58,NH+4 concentration was 4.77 mol · L-1 ,t-CA concentration was 10 g · L-1. The maxima! L-Phe yield was 27.5 g ·L-1 under the optimum conditions.
