摘要
目的 观察缺氧条件下培养的肺微血管内皮细胞 (PMEC)培养液对大鼠主动脉平滑肌细胞(ASMC)的作用。方法 应用倒置显微镜和电镜观察细胞形态 ,同位素放射测定3H 胸腺嘧啶核苷 ( 3H TdR)摄取量 ,流式细胞仪测定细胞的周期和蛋白含量。结果 单纯缺氧可使ASMCG0 G1 期数目增多。3H TdR摄取量降低 ,蛋白质含量减少 ;缺氧的PMEC培养液能促进大鼠ASMC从G0 G1 期进入S期 ,3H TdR含量升高 ,使蛋白质含量进一步降低。结论 缺氧可能刺激微血管的内皮细胞分泌某些活性物质 ,进而促进动脉平滑肌细胞增生。
Objective To observe the effect of supernant of pulmonary microvascular endothelial cell cultured medium (SPMECCM) on rat aortic smooth muscle cells (ASMC) in hypoxia condition. Methods The morphology of ASMC was observed with inverted light microscope and electron microscope. 3H TdR uptake was tested with isotope labelled method. The cell cycle and protein contents were analyzed with flow cytometry (FCM). Results In simple hypoxia group, ASMC in G 0/G 1 phase were increased, but 3H TdR uptake and protein contents were decreased. While in hypoxics SPMECCM treated group, more ASMC grow from G 0/G 1 phase to S phase. 3H TdR uptake was elevated but protein contents were reduced. Conclusion Hypoxia might stimulate microvascular endothelial cells to secrete some growth factors and enhance the proliferation of ASMC.
