摘要
目的 观察心梗愈后心力衰竭大鼠心肌AE3mRNA及其蛋白表达的变化 ,探讨其在心力衰竭中的病理生理意义。方法 结扎大鼠冠状动脉 ,造成心梗愈后心力衰竭模型 ,应用原位杂交及免疫组化技术 ,检测心肌AE3mRNA及其蛋白表达。结果 心衰组心肌AE3mRNA及其蛋白表达显著增加 ,其IOD值明显高于假手术组和正常对照组 (P <0 .0 1)。结论 心力衰竭时心肌AE3mRNA及其蛋白表达显著增加是一种代偿反应 ,但可导致细胞内酸中毒及细胞内Ca2 + 超负荷 ,使心衰加重。
Objective To investigate expressions of mRNAs and proteins of myocardial anion exchanger 3 in rats with congestive heart failure after recovered from myocadial infarction, and explore its pathophysiological significance in cardiac failure. Methods The model of congestive heart failure recovered from myocardial infarction in rats was established by ligating coronary artery. The expressions of mRNAs and proteins of myocardial anion exchanger 3 were determined with in situ hybridization and immunohistochemical method respectively. Results Expressions of mRNAs and proteins of myocardial anion exchanger 3 increased significantly. Integral optimal densities of images from rats with congestive heart failure were higher than sham operation and normal control rats(P<0.01). Conclusion The increase in the expressions of mRNAs and proteins of myocardial anion exchanger 3 in rats with congestive heart failure is a compensatory effect, which may lead to intracellular acidosis and Ca 2+ overload and exacerbates the progression of congestive heart failure.
