斯氏肺吸虫和华支睾吸虫基因组多态DNA的初步分析 被引量:9

摘要 目的 比较分析斯氏肺吸虫和华支睾吸虫基因组多态DNA的多态性和探讨两吸虫间的相似基因。方法 应用随机扩增多态性DNA(RandomamplifiedpolymorphicDNA ,RAPD)技术对斯氏肺吸虫和华支睾吸虫的基因组多态DNA进行扩增 ,扩增产物经琼脂糖凝胶电泳后分析结果。结果 引物P1和P2扩增产生斯氏肺吸虫和华支睾吸虫的成虫DNA图谱 ,而P3引物未扩增出产物。斯氏肺吸虫和华支睾吸虫的P1扩增片段均不相同 ,而P2引物对两吸虫的扩增产物有 2条相同的片段 ,分别为 40 0、10 0bp。结论 RAPD结果显示斯氏肺吸虫和华支睾吸虫为 2种独立的吸虫虫种 ,由于遗传的亲缘关系存在相同的基因 ,这些相同基因是否表达共同的抗原组分 ,并导致血清学诊断的交叉反应尚待进一步研究。 Objective To analyze the polymorphism of the genomic DNA extracted from Paragonimus skrjabini and of that from Clonorchis sinensis and explore the similarity of their genes. Methods The genomic DNA extracted from the 2 trematodes was amplified with random amplified polymorphic DNA (RAPD) and then was carried on agarose gel electrophoresis. Results DNA fingerprints of the 2 trematodes were obtained through the primer P1 and P2 but not through the primer P3. With the primer P1, the amplified fragments from the 2 trematodes were quite different, while with P2, 2 similar bands were found with 400 bp and 100 bp respectively. Conclusion Our results indicate that though Paragonimus skrjabini and Clonorchis sinensis were independent species, similar genes exist because of their close genetic relation. It should be further investigated whether these similar genes express common antigens resulting in cross-reaction in serodiagnosis.
