摘要
目的:通过观察龟鹿二仙胶及其拆方对SD大鼠膝关节软骨退变的影响,探讨龟鹿二仙胶的配伍机制。方法:切除SD大鼠双侧卵巢、双膝内侧副韧带和前交叉韧带、实验跑台跑步4周,制作退化膝骨关节炎模型。中药灌胃2、4、8周后放射免疫法检测血清雌二醇(E2)、免疫组化检测Bax、Bcl-2、白介素1β(IL-1β)、基质金属蛋白酶-13(MMP-13)、Ⅱ型胶原(Collagen-Ⅱ)的表达。结果:8周时龟鹿参组可显著提高大鼠血清E2水平,龟鹿杞组可显著抑制Bax、促进Bcl-2表达,龟鹿参组可显著地抑制软骨细胞产生IL-1β和MMP-13、提高Collagen-Ⅱ的表达,其效果均与龟鹿二仙胶作用相同。结论:龟鹿配伍人参可提高血清E2水平、抑制关节软骨产生炎症、促进软骨细胞Collagen-Ⅱ合成;龟鹿配伍枸杞可以抑制软骨细胞凋亡。
Objective: To observe the mechanism of Guilu Erxian Glue compatibility by detecting the effect of preventing SD rat's knee articular cartilage regression by Guilu Erxian Glue and its seperates. Methods: To prepare a model of degenerating knee osteoarthritis by extirpating SD rat's both ovaries, knee medial collateral ligament and anterior cruciate ligament, and run for 4 weeks. Guilu Erxian Glue or its separate were intragastric administrated for 2, 4, 8 weeks. Then radio-immunity was used to detect serum estradiol, and immunohistochemistry was applied to detect the expression of Bax, Bcl-2, IL-1β, MMP-13, Collagen- Ⅱ. Results: At 8 week, Colla carapacis et plastri testudinis, colla comus cervi and ginseng can significantly raise SD rat's serum estradiol and the expression of Collagen- Ⅱ, significantly inhibit the expression of IL-1β and MMP-13. CoUa carapacis et plastri testudinis, colla cornus cervi and medlar can significantly inhibit Bax, raising Bcl-2 expression. Conclusion: Colla carapacis et plastri testudinis, colla cornus cervi and ginseng can raise serum estradiol, inhibit inflammation produce by arthrodial cartilage, promote chondrocyte composite Collagen- Ⅱ. Colla carapaciset plastri testudinis, colla cornus cervi and medlar can prevent chondrocyte from apoptosis.
