菟丝子黄酮对雷公藤多苷所致生殖损伤的雄性幼鼠睾丸组织中表皮生长因子表达的影响 被引量:18

Effects of GTW on expression of EGF and the intervention effect of Tusizi flavones in male juvenile rats

摘要 目的:研究雷公藤多苷(GTW)对幼年大鼠睾丸组织表皮生长因子(EGF)表达的影响以及菟丝子黄酮的干预作用。方法:3周龄SD雄性幼鼠48只,随机分为4组:空白组,多苷组,六味组(六味地黄丸+GTW),黄酮组(菟丝子黄酮+GTW),持续给药12周后,应用原位杂交及Western Blot法检测各组动物睾丸组织EGF mRNA及EGF蛋白表达。结果:GTW可以明显降低EGF mRNA和蛋白的表达,多苷组与空白组比较有显著性差异(P<0.05);菟丝子黄酮可以明显升高EGF mRNA和蛋白的表达,黄酮组与多苷组比较有显著性差异(P<0.01)。结论:GTW对幼年大鼠睾丸组织EGF mRNA及EGF蛋白表达有一定的抑制作用,菟丝子黄酮能够提高EGFmRNA及EGF蛋白表达量而起到保护GTW所致的雄性幼鼠生殖损伤作用。 Objective: To study the effect of GTW on expression of EGF and the intervention effect of Tusizi flavones in male juvenile rats. Methods: The 48 male juvenile SD rats of 3-weeks old were divided into 4 groups randomly: normal group, GTW group, Liuwei-Dihuang-Wan and GTW group (Liuwei group), Tusizi flavones combined GTW group (flavones group); After 12 weeks of intragastric administration, the expression of EGF mRNA in testis was detected with in situ hybridization technique, the expression of EGF protein in testis was detected with Western Blot. Results: The mean OD value of EGF mRNA and EGF protein expression in testicle tissue in GTW group was remarkably lower than it was in normal group, with significant difference (P〈0.05); The mean OD value of EGF mRNA and EGF protein expression in testicle tissue in flavones group was remarkably higher than it is in GTW group (P〈0.01). Conclusion: We presume that the GTW can decrease EGF mRNA and EGF protein expression in male juvenile rats, which was improved by Tusizi flavones.
