摘要
建立一种合理、准确、快速的体外代谢致毒预测模型,在临床前预测化合物及其代谢物的毒性对于新药研发具有非常重要的意义。该文主要综述了新型的预测方法(转基因肝细胞、共培养技术、3D培养法、高通量代谢装置以及代谢灌流模型等),这些方法都已成功的应用于相关代谢物的毒性预测,为药理、毒理研究提供新思路。
It is extremely important to predict chemical metabolic toxicity in preelinical stage for drug discovery. Thus, this paper mainly reviews some novel, reasonable, accurate and fast pre- dictive models in vitro, including transgenes cells, independent discrete multiple organ co-culture, 3D cell culture, high throughoutput metabolic device, vide fresh inspiron for search.
