摘要
目的从鬼针草中提取抗肿瘤活性成分并确定其结构,测试其对肿瘤细胞株的增殖抑制作用。方法以HeLa细胞为活性成分体外筛选模型,通过HPLC等方法分离提取抗肿瘤活性成分;通过波谱分析技术确定其化学结构;MTT法测试活性成分对多个肿瘤细胞株的增殖抑制作用。结果提取到的鬼针草抗肿瘤活性成分为矢车菊黄素,其对HeLa、U2-OS、A549、MCF-7、HepG-2细胞株的半数抑制浓度(IC50)分别为0.18,1.52,0.67,0.85,0.43μM。结论首次从鬼针草中提取到矢车菊黄素,该化合物具有较强的抗肿瘤细胞增殖作用。
OBJECTIVE To obtain the antitumor compound from Bidens pilosa Linn.,determine its structure and investigate its antiproliferative effect on human tumor cells.METHODS Extraction and isolation of antitumor compound were performed by HPLC with the HeLa cells as the in vitro screening model.Spectrum analysis was used to determine the structure of antitumor compound and MTT assays were performed to investigate its antiproliferative effect on human tumor cells.RESUTLS Bioactivity-guided isolation of antiproliferative compound from Bidens pilosa Linn.resulted in the obtaining of centaureidin,which IC50 values on HeLa,U2-OS,A549,MCF-7,and HepG-2 cells were 0.18,1.52,0.67,0.85,0.43μM respectively.CONCLUSION It is the first time to obtain centaureidin from Bidens pilosa Linn.,which exerts high antiproliferative activity on human tumor cells.
