摘要
[目的]建立一种萃取分离纯化食用色素的新方法。[方法]研究在聚乙二醇(PEG)/硫酸铵双水相体系中,PEG浓度、(NH4)2SO4用量、溶液酸碱度等因素对黑糯玉米色素萃取分离纯化的影响,并利用响应面分析法确定了最佳分离纯化条件。[结果]研究表明,PEG浓度20%、(NH4)2SO4用量为6 g、溶液pH为3.5时,黑糯玉米色素萃取率最高,分离纯化的效果最好。[结论]双水相体系可用于分离纯化黑糯玉米色素。
[Objective]The aim was to establishe an new extract method for the separation and purification of food pigment. [Method]The separation behavior of black glutinous corncob pigment in two-phase aqueous system of Water-PEG in the presence of ammonium sulphate were investigated. The experiments about the effects of different concentrations of PEG, different amount of (NH4)2SO4 and different acidity were performed, and then the separation and purification condition was optimized by response surface analysis. [Result]The result showed that the concentrations of PEG was 20% and the amount of (NH4)2SO4 was 6 g and the pH = 3.5, the extraction rate reached the highest and the effect of the separation and purification was the best. [Conclusion]Aqueous two-phase system could be used to separate and purificate the pigment in black glutinous corncob.
