银杏叶提取物对肝纤维化大鼠肝脏血小板衍生生长因子表达的影响 被引量:3

摘要 目的:观察银杏叶提取物(GbE)对大鼠肝组织PDGF表达的影响。方法:采用CCL4建立大鼠肝纤维化,同时以50、100、200mg(/kg.d)的GbE干预,以复方鳖甲软肝片为对照,连续6周后取肝组织,采用RT-PCR法检测肝组织PDGFmRNA表达,免疫组化法测定肝组织PDGF蛋白的表达。结果:模型大鼠肝组织PDGFmRNA和蛋白表达较正常组明显增高(P<0.01),GbE和复方鳖甲软肝片干预后大鼠肝组织PDGFmRNA和蛋白表达较模型组显著下降(P<0.05,P<0.01)。结论:GbE可能通过降低肝组织PDGF的表达,抑制CCl4诱导的大鼠肝纤维化的进展。 Objective: To investigate the effects of ginkbo biloba extract (GbE) on the expression of platelet-derived growth factor (PDGF) in rat liver tissues with hepatic fibrosis induced by carbon tetrachloride (CCL). Methods: Rat models of hepatic fibrosis were induced by CCL and were treated with GbE of 50, 100, and 200 mg/ kg. d, respectively, for 6 weeks. Biejia Ruangan tablets served as control treatment. At the end of the 6th week, all the rats were painless killed. The transcription of PDGF mRNA in the livers of these rats was analyzed by RT-PCR and the expression of PDGF protein was detected by immunohistochemistry. Results: The expressions of PDGF mRNA and protein were remarkably higher in rat livers with hepatic fibrosis than those in normal ones (P〈0.01), while both GbE and Biejia Ruangan tablets significantly reduced the expressions of PDGF at both levels when compared with the model ones (P〈0.05, P〈0.01). Conclusions: CCL-induced hepatic fibrosis may be inhibited by GbE through reducing the expression of PDGF in rat livers.
