摘要
采用超高效液相-电喷雾质谱法(UPLC-ESI/MS/MS)和超高效液相-大气压化学电离质谱法(UPLC-APCI/MS/MS),分析烟叶中的植物甾醇。使用Waters Acquity UPLC BEH C18色谱柱,以甲醇、水为流动相梯度洗脱,甾醇和烟叶中其它成分分离良好。在ESI源和APCI源正离子模式下获得了甾醇离子碎片,胡萝卜甙、胆甾醇、β-谷甾醇和豆甾醇等不饱和甾醇,脱水[M+H-H2O]+是响应最强的离子片段。通过对烟叶中甾醇的ESI/MS/MS和AP-CI/MS/MS的离子流图进行比较,发现相对于ESI来说,APCI源选择性较好,基质干扰小,说明APCI源更适合烟叶中植物甾醇的定量分析。
A new method based on ultra performance liquid chromatography (UPLC), electrospray mass spectrometry (ESI/MS/MS) and atmospheric pressure chemical ionization mass spectrometry (APCI/ MS/MS) has been developed for the analysis of sterols in tobacco leaves. Sterols and other components in tobacco leaves were separated on an Waters Acquity UPLC BEH C18 column,with methanol and water as mobile phase gradient elution. In ESI and APCI positive ion mode,dehydration [ M + H - H2O] + are the strongest sterol ion fragments, daucosterol,/3-sitosterol and stigmasterol. By comparing ESI/MS/MS and APCL/MS/MS for the analysis of sterols in extract of tobacco leaf,it is found that APCI mode is more selective than ESI mode, so APCI mode is more suitable for sterol quantitative analysis in tobacco.
