摘要
目的 探讨皮肤光老化的发生机理 ,并建立细胞水平的光老化模型。方法 应用正常人来源的单层培养皮肤成纤维细胞 ,长波紫外线照射 2天后 ,用MTT法检测其细胞增殖情况 ,3 H -脯氨酸检测其细胞胶原合成情况。结果 照射组与非照射组细胞增殖差异无显著性意义 (P >0 .0 5 ) ,而照射组细胞胶原合成比非照射组显著降低 (P <0 .0 0 1)。结论 在体外培养情况下 ,短期、小剂量长波紫外线对人成纤维细胞胶原生物合成有显著的抑制作用。
Objective To investigate the mechanism of photoaging and to provide a cellular photoaged model.Methods Fibroblasts monolayers from nomal human skin cultured in vitro were irradiated by UVA light for two days.The fibroblasts' proliferation was measured by MTT method,and the collagen of the cell biosynthesis by 3H proline in corporation.Results There was no statistical difference between the UVA exposed cells and the UVA unexposed cells in proliferation( P >0.05);the collagen of UVA exposed cells biosynthesis was decreased than that of unexposed cells( P <0.001).Conclusion UVA can reduce the collagen of fibroblasts' biosynthesis in vitro.
