摘要
目的 :通过立体定向在 SD大鼠右尾状核区接种 C6 细胞 ,建立类似于人脑胶质瘤的动物模型。方法 :SD大鼠在立体定向条件下 ,左右尾状核区接种 1× 10 6 个 C6 胶质瘤细胞 ,接种后观察实验大鼠的生成状态 ,分别于接种后 1,2 ,3周时进行 MRI检查。在实验三周时解剖标本 ,做组织病理学和 GFAP免疫组化检查。结果 :该方法建立的胶质瘤动物模型在组织病理学上接近人脑胶质瘤 ,而且具有颅内生长稳定 ,成瘤率高 ,没有颅外种植生长 ,实验周期短 ,重复性好。结论 :该大鼠脑胶质瘤动物模型能够满足胶质瘤实验治疗研究的需要 ,而且在细胞接种后 1~ 2周 ,为实验治疗较好的时机。
Objectiv:In order to understand factors that modulate the growth of glioma and to develop treatments that inhibit gliomas.Methods:We established rat cerebral glioma model by implanting 1×10 6 C 6 glioma cell into rat right caudate nucleus with stereotactic technique.Tesults:Rats had obvious intracranial hypertension at 2 weeks after implantation.Rats were in severe danger at 3 weeks after implantation.The death rate was 30%.Intracranial tumors could be detected at 1 week after implantation.Diameter of tumors was almost 2×3 mm at 1 week,5×6 mm at 2 weeks 7×9 mm at 3 weeks.After 3 weeks, all rats were sacrificed.Tumors can be confirmed being belong to glial tumor cells because of strong postitive staining of glial fibrillary acidic protein (GFAP) with immunohistochemistry.Rat C 6 gliomas resembled histopathological key features of human glioblastoma.The stereotactic implantation of tumor into the rat caudate nucleus resulted in a more reliant and reproducible tumor model,with 100% yield of intracerebral tumor as well as marked reduction in extracranial extension.Conclusion:This glioma model can satisfy the need of experimental treatment.The time of treatment would be selected at 1-2 weeks after implantation.
