抗痛风胶囊改善急性痛风性关节炎大鼠关节病理学变化

Anti-gout capsule improves the pathological changes in joint of acute gouty arthritis rats

目的探讨抗痛风胶囊对急性痛风性关节炎大鼠关节病理学变化的影响。方法采用尿酸钠混悬液关节腔内注射诱导大鼠急性痛风性关节炎模型,连续用药治疗5 d,肉眼观察大鼠足踝关节肿胀程度,HE染色观察踝关节软骨组织的病理变化。结果与模型组比较,抗痛风胶囊和阳性药对照组AGA大鼠于相应药物治疗后3～5 d,右足踝关节肿胀明显减轻。HE染色后显微镜下观察结果显示,与空白组比较,模型组大鼠踝关节软骨组织表面较不整齐,基质染色不均,软骨细胞排列层次紊乱,细胞肿胀明显,有较多的炎细胞浸润,软骨下骨与髓腔也出现炎细胞浸润,充血明显。抗痛风胶囊低剂量和高剂量组、阳性药物组均明显改善关节软骨组织的充血、水肿和炎细胞浸润等病理变化。结论抗痛风胶囊可以明显减轻AGA大鼠关节肿胀程度,改善软骨组织的病理性变化。

Objective To explore the effects of anti - gout capsule on pathological changes in arhcular cartilage tissues of acute gouty arthritis （AGA） rats. Methods Fifty male SD rates were randomly divid- ed into the control, model, positive control （ cochicine ） and anti - gout capsule （ low dose and high dose） groups with 10 rats in each group. AGA model was established by the injection of sodium urate into the joint cavity. Rats in the positive control （cochicine） and anti -gout capsule （low dose and high dose） groups were fed by garage with cochicine （0.8 mg/kg） and anti - gout capsule （0.3 and 1.2 g/ kg） once a day for 5 days. Rats were sacrificed 2 h after the last intragastric administration. Rat ankle joint swelling was observed by the naked eyes and the pathological changes of rat articular cartilage were analyzed by hematoxylin - eosin （HE） staining. Results Compared with the model group, AGA rat an- kle joint swelling in positive control and anti - gout capsule groups was significantly attenuated. HE stai- ning assay showed that compared with the control group, the irregular surface of rat ankle joint cartilage, the uneven dyeing of matrix, the disarrangement of cartilage cells, obvious cell edema and a large num- ber of inflammatory cells infiltration were presented in rat cartilage and marrow chamber of model group, which was significantly attenuated by cochinice and anti -gout capsule （low dose and high dose） treatement. Conclusion The anti - gout capsule could relieve the cartilage damage in AGA rats.
