婆婆针愈伤组织及试管苗培养研究

摘要 为保护野生资源,满足栽培需求,以婆婆针嫩茎为材料,进行了愈伤组织培养和分化培养、不定芽生根和生根继代繁殖培养以及试管苗移栽和定植的研究。结果表明:MS+蔗糖30 g/L+BA 0.4 mg/L+2,4-D 1.8 mg/L是嫩茎段愈伤组织诱导培养和继代繁殖培养的理想培养基;1/2MS+AgNO31.0 mg/L+ZT 0.4 mg/L+NAA 0.1 mg/L是嫩茎愈伤组织分化不定芽培养的理想培养基。White+ABT 2号1.4 mg/L+蔗糖12 g/L+IAA 0.3 mg/L是不定芽生根培养和试管苗生根继代繁殖培养的理想培养基;试管苗移栽和定植容易成活;定植的试管苗保持了野生婆婆针的所有植物学性状。 In order to preserve the wild resources and meet the cultivated needs of people,taking the tender stems of Cichorium intybus as material,research was conducted by callus tissue culture,inducement differentiation,rooting of adventitious buds,rooting multiplication of tube seedlings, transplantation of tube seedlings and stable planting.The results showed that MS+ sucrose 30 g/L+BA 0.4 mg/L+2,4-D 1.8 mg/L was the ideal medium for callus inducement and callus multiplication.l/2 MS+AgNO3 1.0 rag/L+ ZT 0.4mg/L+NAA 0.1 mg/L was the optimum medium for callus differentiation cultivation and adventitious buds cultivation.White+ABT 2 1.4 mg/L+sucrose 12 g/L+IAA 0.3 mg/L was the ideal medium for rooting of adventitious buds and rooting multiplication of tube seedlings.Transplantation of tube seedlings and stable planting were easy ,and the tube seedlings stable planted maintained all botany characteristics of the wild Cichorium intybus.
