摘要
目的 :探讨急性白血病患者 WT1基因与 L RP基因表达的关系。方法 :应用逆转录 -聚合酶链反应(RT- PCR)法检测 73例急性白血病患者及 2 3例正常人的 WT1及 L RP基因的表达。结果 :73例急性白血病患者中 41例 WT1基因表达阳性 ,2 5例 L RP基因表达阳性 ;2 3例正常人中 WT1基因表达均阴性 ,仅 1例 L RP基因表达阳性。 WT1基因表达阳性患者的完全缓解 (CR)率低于表达阴性者 (分别为 5 3.7%和 84.4% ,P <0 .0 1) ;L RP基因表达阳性患者的 CR率亦低于表达阴性者 (分别为 5 2 .0 %和 75 .0 % ,P <0 .0 5 )。 WT1表达阳性患者的L RP阳性率高于 WT1表达阴性者 (分别为 46 .3%和 18.8% ,P <0 .0 5 )。结论 :WT1基因的表达和 (或 ) L RP基因的表达与急性白血病患者的化疗效果及预后密切相关 ,是影响急性白血病患者 CR率的重要危险因素 ,且二者有一定的相关性。
Objective:To evaluate the relationship between WT1 and LRP genes expression and their prognostic value in acute leukemia.Methods:Expressions of WT1 and LRP genes were measured in 73 patients with acute leukemia and 23 normal controls by RT PCR method.Results:Expression of WT1 gene was found in 41 of 73 acute leukemia patients and none of 23 normal controls. Expression of LRP gene was found in 25 of 73 acute leukemia patients and 1 of 23 normal controls. Complete remission rate (53.7%) was lower in WT1 positive patients than in those ( 84.4 %,P< 0.01 ) WT1 negative patients. Complete remission rate ( 52.0 %) was also lower in LRP positive patients than in those ( 75.0 %, P< 0.05 ) LRP negative patients. The rate of LRP expression was higher in patients with WT1 expression ( 46.3 %) than in those without WT1 expression ( 18.8 %,P< 0.05 ). Conclusion:The expression of WT1 and/or LRP gene can predict the treatment outcome and prognosis in acute leukemia patients. Expressions of WT1 and LRP are important risk factors affecting CR rate of acute leukemia patients.
