摘要
目的建立荷叶配方颗粒高效液相色谱指纹图谱,为其质量控制提供科学、合理的方法。方法采用高效液相色谱法,以Merck HibarPurospherSTAR RP-18e柱(4.6 mm×250 mm,5μm)为色谱柱,甲醇、0.4%(体积分数)H3PO4-0.2%(体积分数)三乙胺为流动相梯度洗脱。检测波长为270 nm;流速为0.8 mL.min-1;柱温为25℃。结果建立了荷叶配方颗粒的指纹图谱,标示了13个共有峰,并对10批荷叶配方颗粒HPLC指纹图谱进行了相似度评价,各荷叶配方颗粒样品指纹图谱与对照指纹图谱相似度均在0.90以上,可以用于荷叶配方颗粒的定性鉴别。结论该方法简单、准确、重复性好,为有效地控制与评价荷叶配方颗粒的质量提供了可靠的方法。
Objective To establish a method of fingerprint analysis on Nelumbo nucifera Gaertn by HPLC for the quality control.Methods The HPLC chromatographic fingerprints were obtained with Merck HibarPurospherSTAR RP-18e column(4.6 mm×250 mm,5 μm) by gradient elution solvent system composed of methanol-water(0.4% phosphoric acid and 0.2% triethylamine).The wavelength was set at 270 nm;the flow rate was 0.8 mL·min-1 and the column temperature was maintained at 25 ℃.Results Thirteen main common peaks were selected in the fingerprint peaks.Good similarities with correlation coefficients higher than 0.90 were found in fingerprints between different samples of Nelumbo nucifera Gaertn and the standard fingerprint.Conclusion The method is simple and accurate with good reproducibility,which may be used for the quality control of Nelumbo nucifera Gaertn.
