摘要
目的研究四君子汤对大黄脾虚模型小鼠机能状态,脾脏、胸腺指数及病理形态学的影响,探讨四君子汤治疗脾虚的作用机制。方法大黄水灌胃建立小鼠脾虚模型。小鼠随机分为对照组、脾虚模型组、低剂量治疗组(0.5 g/mL四君子汤)和高剂量治疗组(1 g/mL四君子汤),每组15只,给予相应干预措施。干预7 d后分别观察小鼠体质量、体温,测量脾脏、胸腺湿重并计算相应脏器指数;结合显微镜下观察脾脏、胸腺的病理形态学变化进行分析。结果四君子汤各剂量治疗组的体温、体质量增长及脾脏指数、胸腺指数均显著高于脾虚模型组;四君子汤各剂量治疗组小鼠脾脏、胸腺萎缩现象得到改善,脾脏淋巴细胞聚集、中央动脉闭塞与脾虚模型组相比均有明显改善,红髓区巨核细胞明显增多;胸腺淋巴细胞数量亦较脾虚模型组多,以高剂量治疗组更明显。结论四君子汤能有效促进脾虚模型小鼠的恢复,其机制可能与促进脾脏和胸腺组织T淋巴细胞修复有关。
Objective To study the effect and mechanism of Sijunzi-tang on rhubarb-induced spleen deficiency in mice with basic functional state,thymus index and pathomorphology.Methods The mouse model of spleen deficiency was established with rhubarb treatment.Mice were randomly divided into four groups,including the negative control group,positive control group,low(0.5 g/mL) and high(1 g/mL) Sijunzi-tang groups.The changes of body weight,temperature,wet weight and organ index of the spleen and thymus were determined at day 7.The pathomorphological changes of spleen and thymus were also checked under microscopy.Results Treatment with Sijunzi-tang significantly improved body weight,temperature,wet weight and organ index of the spleen and thymus compared with the positive control group.Sijunzi-tang also obviously ameliorated the atrophy of spleen and thymus,spleen lymphocytes gathering and central artery occlusion.Furthermore,Sijunzi-tang,especially in the high dosage,enhanced red marrow megakaryocytes and thymus lymphocytes.Conclusion Sijunti-tang could effectively promote the rehabilitation of rhubarb-induced spleen deficiency in mice,and its mechanism may be related to upregulation of the activity of T lymphocytes in spleen and thymus.
