六味地黄丸含药血清减轻KCl诱导的PC12细胞损伤

摘要 目的观察六味地黄丸含药血清对KCl诱导PC12细胞损伤的影响。方法取对数生长期的PC12细胞,分为对照组、模型组、六味地黄丸含药血清组,采用KCl诱导PC12细胞损伤模型,观察细胞的形态和成长状况,MTT法测定细胞活性及乳酸脱氢酶(LDH)活力,用RT-PCR法检测细胞CREB mRNA表达,SABC免疫组化法检测细胞CREB的表达。结果模型组细胞上清液中LDH活力显著增高于对照组(P<0.01),细胞的CREB mRNA及蛋白表达显著减少(P<0.01);六味地黄丸含药血清组细胞上清液中LDH活力较模型组显著降低(P<0.01),细胞CREBmRNA及蛋白表达显著升高(P<0.01)。结论六味地黄丸含药血清可减轻KCl诱导的PC12细胞损伤。 Objective To investigate the effect of the drug-containing serum of Liu Wei Di Huang Wan on PC12 cells which were damaged by KC1. Methods We established the PC12 cell model induced by KC1 which was di vided into three groups: control group, model group, Liu Wei Di Huang Wan containing serum group. The mor phology of cells in each group were observed with inverted phase contrast microscope. The proliferation of PC12 cells was detected by MTT assay and the concentration of LDH in PC12 cells were analysed. The level of CREB mRNA was analysed by RT-PCR. The expression of CREB was analysed by SABC-immunohistochemistry stain. Re sults The activity of LDH released from cells was increased and the level of CREB was decreased in the model group (P 〈0. 01 )compared with the control group. It significantly improved the activity of cells and the level of CREB mRNA in Liu Wei Di Huang Wan serum group as compared with the model group (P 〈0. 01 ). Conclusions The drug-containing serum with Liu Wei Di Huang Wan can reduce the damage of PC12 cells induced by KC1.
