摘要
目的探讨姜黄素对CD34+人白血病细胞株KG1a增殖和凋亡的影响。方法分别采用0、40、60、80μmol/L的姜黄素处理KG1a细胞,倒置显微镜下观察细胞形态学改变;采用Western blot法测定细胞外信号调节激酶(ERK)、磷酸化ERK(p-ERK)蛋白表达,以及细胞周期相关蛋白P21和P27表达。结果姜黄素作用KG1a细胞48 h后可见细胞皱缩、细胞数减少,呈现显著凋亡形态学改变;姜黄素可抑制p-ERK蛋白表达,对ERK表达水平无明显影响。姜黄素可上调P21和P27蛋白水平。结论姜黄素可抑制KG1a细胞增殖,诱导其凋亡,其机制可能为通过抑制p-ERK表达、上调P21和P27蛋白水平实现的。
Objective To explore the cytotoxic and pro-apoptotic efficacy of eurcumin on CD346 acute myeloid leukemia cells. Methods KG1a cells were treated with different concentrations of curcumin (0,40,60,80 μmol/L).The morphological changes were detected by inverted microscope. The effect of curcumin on the protein expressions of ERK, p-ERK, P21 and 1727 were detected by Western blot. Results The results showed that curcumin induced morphological changes such as cell shrinkage, cell number reduction, which was typical characteristic of apoptosis. Western blot assay showed that curcumin caused significantly down-regulation of p-ERK protein levels, up-regulation of P21 and P27 protein levels. Conclusion Curcumin suppresses cell growth and induces apoptosis in KGla cells. Suppression of p-ERK, up-regulation of P21 and P27 could contribute to curcumin-induced apoptosis in KGla cells.
