胰岛素对人食管鳞癌细胞化疗增敏的影响及作用机制 被引量:3

摘要 目的探讨胰岛素对人食管鳞癌细胞株EC9706化疗敏感性的影响及机制。方法顺铂处理食管鳞癌EC9706细胞,MTT法检测细胞活力,探寻活细胞数量达到平台期的时间T1;加入胰岛素继续培养,探寻活细胞数再次达到平台期的时间T2。用Annexin V-FITC+PI和MDC进行细胞染色,荧光显微镜下定性观察细胞荧光情况;流式细胞术定量检测细胞凋亡、测定MDC荧光强度。结果 (1)顺铂可以诱导EC9706细胞的自噬和凋亡。(2)胰岛素可以增强顺铂对肿瘤细胞的杀伤。(3)T1平台期细胞凋亡比例低于T2平台期(P<0.05),但T1平台期细胞MDC平均荧光强度明显高于T2平台期(P<0.05)。结论在顺铂作用后期,胰岛素通过抑制自噬引起细胞凋亡增加,实现其化疗增敏作用。
