摘要
目的探讨胎儿酒精综合征(fetal alcohol syndrome,FAS)中第21天子代动物中枢神经细胞骨架蛋白的变化。方法用C57BL/6J小鼠建立FAS动物模型,利用免疫组化方法测定高分子量神经丝(high molecular weightneurofilament,NF-H)、中分子量神经丝(medium molecular weight neurofilament,NF-M)、低分子量神经丝(low molecularweight neurofilament,NF-L)、微丝蛋白β-actin、微管蛋白β-tubulin在第21天仔鼠大脑皮质、海马区和小脑中含量的变化。结果酒精暴露组第21天仔鼠,NF-H在大脑皮质和海马区含量均明显降低(P<0.01),而在小脑组织中含量未见明显下降;NF-M在大脑皮质、海马区和小脑中含量均明显降低(P<0.05或P<0.01);NF-L在大脑皮质、海马区和小脑中含量均明显降低(P<0.05或P<0.01);β-actin在大脑皮质、海马区和小脑中含量均未见明显降低(P>0.05);β-tubulin在大脑皮质、海马区和小脑中含量均明显降低(P<0.05或P<0.01)。结论中枢神经细胞骨架蛋白在FAS中发生了明显变化,由此推断,其可能是FAS发病的早期靶点。
Objective To study the alterations of cytoskeletal proteins in central nervous system of 21 days postnatal mice in Fetal Alcohol Syndrome (FAS) mouse model. Methods The C57BL/6J mouse was used to establish the FAS mouse model. Immunohistochemistry was used to investigate the alterations of high molecular weight neurofilament ( NF-H), medium molecular weight neurofilament (NF-M), low molecular weight neurofilament (NF-L), [^-actin and [^-tubulin in the cerebral cortex, hippocampus and cerebellum of 21 days postnatal mice, when the pregnant mice were treated with 10% (v/v) ethanol in drinking water. Results Compared with control, the content of NF-H protein was decreased significantly in the cerebral cortex and hippocampus (P 〈 O. 01 ) but no alteration was found in the cerebellum; the contents of NF-M protein, NF-L protein and ^-tubulin protein were decreased significantly in the cerebral cortex, hippocampus and cerebellum ( P 〈 0. 05 or P 〈 0. O1 ) ; no alterations of the content of 13-actin protein were found in the cerebral cortex, hippocampus and cerebellum ( P 〉 O. 05 ). Conclusion Cytoskeleton maybe the beginning target of FAS in view of the significant alterations of the cytoskeletal proteins in central nervous system found in FAS mouse model.
