左卡尼汀联合比索洛尔对心肺复苏大鼠心肌凋亡的影响

Effects of Combination of L-carnitine and Bisoprolol on Myocardial Apoptosis after Cardiopulm Onary Resuscitation in Rats

摘要 目的探讨左卡尼汀联合比索洛尔对心肺复苏大鼠凋亡的影响及其心脏保护作用。方法清洁级雄性SD大鼠48只,随机分为4组(n=12):假手术组(Sham组)、心肺复苏对照组(NS组)、左卡尼汀及比索洛尔联合治疗组(L-B组)及左卡尼汀组(Lc组)。建立大鼠心肺复苏模型,HE法检测复苏后大鼠心肌病理改变,Tunnel法检测复苏后心肌细胞凋亡情况;免疫印迹检测复苏后心肌组织Bax及细胞色素C蛋白表达。结果 L-B组及Lc组与Sham组及NS组相比,心肺复苏后心肌病理变化减轻,凋亡细胞减少;心肌组织线粒体中Bax表达水平明显减少,细胞色素C则相反,其中L-B组更明显。结论左卡尼汀联合比索洛尔,可以通过减少线粒体中Bax表达及增加细胞色素C表达,减轻细胞凋亡,对心肺复苏后大鼠心肌有保护作用。 Objective To investigate the effect of Combination of L - earnitine and bisoprolol on myocardial apotosis after cardiopulmonary resuscitation in rats. Methods Totally 48 SD rats were randomly divided into 4 groups : Sham operation group ( Sham group) ; Cardiopulmonary resuscitation group ( NS group) , L - carnitiue and bisoprolol therapy group ( L - B group) and L - carnitine therapy group (Lc group). HE method was used for the detection of myocardial pathological changes after cardiopulmonary resuscitation in rats, and myocardial apoptosis were detected by Tunnel method, while Bax and Cytochrome C expression in cytoplasm and mitoehondria was detected by Western blotting. Results Compared with NS and Sham group, myocardial pathological changes, apoptotic cells number and the Bax expression in myocardial tissue mitochondria in L - B and Lc group were decreased significantly while the L - B group was more obvious. Conclusion Combination use of levocarnitine and bisoprolol can reduce the expression of Bax, increase cytochrome C expression in mitochondrial, reduce apoptosis, after cardiopulmonary resuscitation in rats, and may have a protective effect.
