摘要
目的探讨线粒体DNA对模拟日光照射诱导凋亡的作用。方法实验分照射组和假照射组,照射组以3.39J/cm2+177mJ/cm2的剂量照射Rho206细胞和143B细胞,假照射组作同样处理但不照光。6h时检测活性氧簇(ROS)产量;16h后观察其细胞形态,检测凋亡率。结果照射组Rho206细胞、143B细胞的凋亡率和ROS的产量与假照射组比较明显上升,差异有统计学意义(P<0.05),照射组Rho206细胞的凋亡率和ROS的产量与143B细胞的凋亡率和ROS的产量比较明显下降(P<0.05),而假照射组两细胞凋亡率和ROS的产量比较差异无统计学意义。结论线粒体DNA缺失细胞对模拟日光照射诱导的凋亡作用的抵抗可能与ROS生成减少有关。
Objective To explore the role of mitochondrial DNA in simulated solar UV-irradiation induced apoptosis. Methods Irradiation groups(IG) was treated with UV of 3.39 J/cm2+177 mJ/cm2(UVA+UVB) dosages irradiation, and the shamed groups(SG) was treated with 0 J/cm2+ 0 mJ/cm2 dosages. 16h later, morphology of apoptosis, apoptosis rates were detected and ROS production were detected after irradiation 6 h. Results Compared to SG, apoptosis was pronounced increase both in Rho and parental cells irradiated with stimulated solar irradiation (P〈0.05). Moreover, apoptosis were marked lower in Rho ceils compared with wild-type cells (P〈0.05) in IG. However, there was no different in apoptosis between two cell lines in SG. Conclusion Mitochondrial DNA-depleted cells may be resistance to simulated solar UV-irradiation triggered apoptosis by decreased ROS yielded.
